Describe the “step-by-step” procedures that you would follow in determining the TCID50 of cytomegalovirus that is known to infect monkey kidney (MK-12) cells. Both the virus stock and MK-12 cells are kept differently at -800C              (20 marks)

a) Briefly describe how you would isolate primary epithelial cells from a pig kidney (10 marks)

b) Write short notes on how you would maintain Vero cells (10 marks)

You have been hired by the Ministry of Health and Social Welfare as a Principal Laboratory Technologists to start a virology cell culture laboratory. Advice your employer on the following:

a) a drawing of the laboratory layout including the placement of major equipment required (10 marks),

b) a list of consumables, supplies and materials required to be purchased and their function (10 marks),
